Ok, so I watched teh first volume of Saiyuki Reload today! Hmm. Here's what I thought of it.... (no spoilers or anything)
First of all, like Emi (and Greg) said, you hafta watch it in Japanese! Ack! ALL teh English voices are different! It's just not right, I say. (It made Davy cry!) But teh Japanese voice cast is teh same, so it's not teh end of teh world. As much as I liked teh dub on teh original, I can listen to Seki Toshihiko all day. (He does Sanzo... and Count D... and mmmmm, Riki..... and Legato!)
In general, teh animation is pretty similar, at least as far as I've seen. There are a few minor design changes; Gojyo got a new outfit, which I actually kind of appreciate. (What was up with those pants, anyway?) Kougaiji probably looks teh most different, but nothing too jarring.
Overall, it does have a bit of a different feel from teh original. They really play up teh Saiyuki 'attitude', both in teh music and teh animation. (teh opening theme sounds a bit like Rage Against teh Machine.) Sort of like, Saiyuki looked around one day and realized, "Hey, I'm super-cool!". It's not a bad thing, though.
teh eps were great, funny and cool and one sad one, and everything that I wuv about Saiyuki. I think once I get used to it a bit, it will be just as great as teh original. Now if only I didn't hafta wait so long for teh next one to come out....
(Oh, and there's also thes short little 'Saiyuki extras' in between each episode...very cute!)

My understanding of Reload's storyline is that its like Gluhen was for Weiss Kreuz. Same characters but different take on teh same story.

It's kinda chronological, but teh animes, manga and drama CD's all seem to sort of go their own way, too. Dunno!

Well nothing can be as different from teh origional as Gluhen from Weiss....But thats cause they didn't wanna pay for teh origional chara designs......
